I bought my RX 330 brand new and has experienced similar problems with it. I have also been left sitting in the middle of the road because the vehicle suddenly stopped, yet the dealer never found out what was wrong with it. I have since had the gasket in rear spoiler broken and the remote opening and falling apart with no warning. The problem with the transmission is also another issue. I am now having problems with the brakes when on the highway, it accelerates before it brakes resulting in multiple near misses. The windshield washer dispenser also freezes in cold weather. I was driving on the beltway and could not see where I was going because I was unable to wash the splashes from passing vehicles off my windshield to see where I was going. Luckily for me it was early morning and the highway was not too busy, coupled with the fact that the route was very familiar to me. I have 18K miles on it and I am thinking of doing away with it.
I have had similar problems with the transmission and Lexus in general. I was looking forward to a positive experience with my first Lexus. Unfortunately, I was extremely disappointed.
